5-Amino-1MQ is a selective, membrane-permeable small-molecule inhibitor of nicotinamide N-methyltransferase (NNMT), an enzyme that depletes the methyl donor SAM in adipose tissue. Blocking NNMT is studied as a way to shift cellular metabolism toward fat oxidation and energy expenditure rather than fat storage.
In a 2018 study, daily 5-Amino-1MQ dosing in diet-induced obese mice reversed high-fat-diet weight gain and improved glucose tolerance over 11 days without reducing food intake. A 2024 study in Scientific Reports found 5-Amino-1MQ sustainably increased voluntary running distance in mice, pointing to effects on muscle performance beyond fat metabolism. All published evidence to date is preclinical; no completed human trials have been published.
